Heat rises. It will be sucked out vent as soon as it enters room when fan is on. It may also suck it out of the register and subsequently other rooms even when the heating is off

Most homes use the fans so infrequently I would would not be to concerned but if they have kids you might suggest a timer so it wont run on for long periods.

It's going to suck out conditioned air no matter where the supply duct is located. I agree with the others who say inconsequential no more than it will be used.
